AAV is known to be the most common vector in gene therapy. During the triple transient transfection system, one transfer plasmid, one helper plasmid (AAssistV™-pHelper) and one Rep/Cap plasmid (AAssistV™-pRC) will be co-transfected into the suspension PowerS™-293 cell line and form AAV.
The ITR regions in AAV plasmids are notoriously unstable. Disruption of ITRs reduces efficiency in viral replication and encapsulation, and increases variability in downstream studies. PowerS™-ITRrs strain is a newly developed strain from ProBio. With proprietary technology, the strain is able to solve the ITR recombination issue during AAV plasmid manufacturing, improving the platform performance.
